چکیده

Ezetimibe is an approved drug for lowering plasma LDL (low-density lipoprotein) level via inhibition of cholesterol absorption. Derivatives ezetimibe reduce inflammatory response and oxidative stress. In the present study, we investigated effect dietary supplementation with in to environmental stressors found that increases resistance stress ultraviolet irradiation. also significantly extended lifespan accompanying reduced fertility, which a common trade-off longevity C. elegans. Cellular reactive oxygen species was increased expression stress-responsive genes, hsp-16.2 sod-3, induced by ezetimibe, suggesting hormetic on lifespan. prevented amyloid beta-induced toxicity completely reversed mortality high-glucose diet. Nuclear localization DAF-16 required prevention enhanced supplementation. Lifespan assay using known long-lived mutants, age-1, clk-1, eat-2, revealed extension specifically overlapped eat-2 are genetic models restriction. Effect worms fed diluted bacteria suggested mimics restriction These findings suggest exhibits anti-oxidative anti-aging effects through hormesis works as dietary-restriction mimetic extension.

Glycogen controls Caenorhabditis elegans lifespan and resistance to oxidative stress

A high-sugar diet has been associated with reduced lifespan in organisms ranging from worms to mammals. However, the mechanisms underlying the harmful effects of glucose are poorly understood. Here we establish a causative relationship between endogenous glucose storage in the form of glycogen, resistance to oxidative stress and organismal aging in Caenorhabditis elegans. We find that glycogen ...

Chlorophyll enhances oxidative stress tolerance in Caenorhabditis elegans and extends its lifespan.

Green vegetables are thought to be responsible for several beneficial properties such as antioxidant, anti-mutagenic, and detoxification activities. It is not known whether these effects are due to chlorophyll which exists in large amounts in many foods or result from other secondary metabolites. In this study, we used the model system Caenorhabditis elegans to investigate the anti-oxidative an...

S-allylmercapto-N-acetylcysteine protects against oxidative stress and extends lifespan in Caenorhabditis elegans

S-allylmercapto-N-acetylcysteine (ASSNAC) was shown in our previous study to activate Nrf2-mediated processes and increase glutathione level and resistance to oxidative stress in cultured endothelial cells. In this study, we explored the antioxidant protective effect of ASSNAC in Caenorhabditis elegans (C. elegans). Treatment of gst-4 reporter strain (CL2166) with increasing concentrations of A...

Minocycline treatment increases resistance to oxidative stress and extends lifespan in Drosophila via FOXO

Minocycline is a semi-synthetic tetracycline derivative antibiotic that has received increasing attention for its non-antibiotic properties, mainly anti-inflammatory, tumor-suppressive, and neuroprotective effects. Drosophila is a widely used genetically tractable model organism for studying organismal aging by virtue of its short lifespan and ease of cultivation. In this study, we examined the...

Loss of NDG-4 extends lifespan and stress resistance in Caenorhabditis elegans

NDG-4 is a predicted transmembrane acyltransferase protein that acts in the distribution of lipophilic factors. Consequently, ndg-4 mutants lay eggs with a pale appearance due to lack of yolk, and they are resistant to sterility caused by dietary supplementation with the long-chain omega-6 polyunsaturated fatty acid dihommogamma-linolenic acid (DGLA). Two other proteins, NRF-5 and NRF-6, a homo...
